ADAMII CD34 requires 4 steps to prepare sample for measurement
The 4 steps are staining - lysis- load- insert.
In order to test real samples, antibody (gray label) and RBC 10x lysis buffer (red label) are required.
| Sample type | ①Staining (µL) | ② Lysis | ③ Loading |
| Fresh/Frozen HPC-A | 20 µL (sample) + 5 µL antibody |
Add 250 µL of 1 x RBC lysis buffer |
25 µL |
| MPB | 20 µL (sample) + 5 µL antibody |
Add 35 µL of 1 x RBC lysis buffer |
|
| Fresh/Frozen Cord Blood | 50 µL (sample) + 5 µL antibody |
Add 200 µL of 1 x RBC lysis buffer |
①Staining
Mix sample and antibody (grey labeled) as the following below.
Please make sure the sample is mixed before performing the staining.
Please see the dilution article before you perform staining.
②Lysis
Lysis must be performed with 1 x RBC lysis buffer.
10x RBC lysis buffer is provided. Dilute with PBS.
Recommend creating 1 mL of 1x RBC lysis buffer. ( 100µL 10x RBC lysis buffer + 900 µL of PBS)
③Loading
Loading is always 25µL regardless of sample.
After loading, wait for 3 minutes and then load chips.
